  1. The Ploidy Analyzer is shipped in a cardboard box that is attached to a wooden pallet with a plastic wrap. Remove the wrap and open the top of the box with a knife or razor blade. Remove foam packing, inserts, bottles and cap assemblies (sheath and waste).

  3. Insert waste level sensor plug into connector labeled "Waste".

  4. To reduce bubbles angle bottle while filling and have sheath run down inside of bottle. Fill sheath bottle to approximately 700ml mark. When inserting probe and pickup tube assembly move the assembly up and down several times to reduce likelihood of trapping a bubble in the sheath line prior to attaching and tightening the cap.

    • Attach waste cap assembly to waste bottle. Secure tightly by hand but do not overtighten and crack the cap. This bottle acts as a vacuum reservoir since it is a vacuum based system.

  5. Insert end of electrical cord below master power switch and check all fluid and sensor connections. Plug the other end of the power cord into suitable wall outlet or PowerVar UPS if available. Plug the keyboard and mouse USB connections into the computer connections located at the bottom left side of the instrument.

  6. Provide support under the left side of the cover when opened so as not to put excessive stress on the hinge support. For support, a box of lens cleaners or chem wipes, or test tube rack can be used.

  7. Upon opening the cover you will notice foam packing on the optical bench, this needs to be removed as part of the setup. Please save and reuse if the instrument will be transported

    • If the instrument will be aligned see the Alignment Procedure for insertion of interlock key, if not the instrument cover can be closed and secured with the screws.

    • Remove the sample tube from the sample port if present.

    • Insert and tighten both long 5mm hex head screws.

  8. Ensure the master switch (located at the rear above the power cord receptacle) is flipped to the ON position. Remove the protective paper if it is present. Turn the instrument on by pushing the power button located at the left rear of top cover (main power switch at rear of instrument needs to be on as well)

    • The system will take a few minutes to completely boot up to the Windows desktop.

  9. Double click on the Cube 16 icon (Cube 15 if it is an older system) to open up the CyView application. Note, some systems may automatically open into the CyView application upon bootup. At the login screen left click in the Login area and type USER, click in the Password area and type Cube1. You can either press the Enter key or left click on Work with CyView.

  10. Select the Prime function by left clicking on the Prime icon and start the Prime sequence by left clicking on the arrow located at the bottom right (labeled as Start the Selected Batch if the mouse is over it.   11. Follow the onscreen instructions and place a 12 x 55 mm tube half filled with the Decontamination Solution (item 04-4010_R) up into the sample port until it clicks. On screen prompts will indicate what needs to be done throughout the Prime process. Proceed accordingly and use a tube half filled with DI H2O at the last step as indicated.

  12. Left click on the Work icon (to the right of the Prime icon) and

    • Left click the Start the Batch arrow to put the instrument in the standard run mode.

    • At this point the instrument is ready for running the calibration (alignment beads). Depending on the configuration you will need to check the alignment for the UV LED, Green Laser or both.
